High-Affinity GHS-R1a Binding
GHRP-2 binds GHS-R1a with higher affinity than GHRP-6, producing larger GH release per microgram dose via the IP3/PKC intracellular cascade in pituitary somatotrophs.
Hypothalamic Ghrelin Mimicry
Central GHS-R1a activation in the hypothalamus produces appetite stimulation through NPY/AgRP neurons, though to a lesser degree than GHRP-6 due to subtle differences in receptor selectivity.
GHRH Synergy
Combined with GHRH analogues, GHRP-2's ghrelin-axis activation produces synergistic GH pulses — the same two-pathway mechanism that makes all ghrelin-class secretagogues more effective when combined with GHRH agonists.
Cortisol and Prolactin Effect
GHRP-2 elevates cortisol and prolactin more than ipamorelin but comparably to GHRP-6. This ACTH activation is dose-dependent and a consideration in extended or high-dose protocols.
