Reactive Oxygen Species Neutralisation
Glutathione (GSH) is the most abundant intracellular antioxidant, present at millimolar concentrations in hepatocytes and micromolar concentrations in most other cells. It directly neutralises hydrogen peroxide, lipid peroxides, and hydroxyl radicals via glutathione peroxidase, and regenerates other antioxidants (vitamins C and E) from their oxidised forms.
Liver Phase II Detoxification
Glutathione conjugation is the primary Phase II detoxification reaction in the liver. GSH-S-transferases conjugate glutathione to reactive electrophiles — including heavy metals, medication metabolites, and environmental toxins — converting them to water-soluble forms for urinary excretion. Adequate hepatic glutathione is essential for effective toxin clearance.
Melanogenesis Inhibition
Glutathione inhibits tyrosinase — the rate-limiting enzyme in melanin synthesis — via copper chelation at the active site and substrate competition. This reduces melanin production in melanocytes, gradually lightening existing hyperpigmentation and preventing new dark spot formation. The skin-brightening effect is most pronounced with injectable rather than oral supplementation.
Mitochondrial Protection
Mitochondria generate the majority of cellular ROS as a byproduct of ATP production. Mitochondrial glutathione (mGSH) is the primary defence against this oxidative burden. Age-related mGSH depletion correlates with mitochondrial dysfunction, and repletion of mGSH protects the electron transport chain from oxidative damage — a mechanism relevant to energy and longevity.
