FOXO4-p53 Complex Disruption
Senescent cells survive by sequestering p53 in the cytoplasm via interaction with FOXO4, preventing p53 from entering the nucleus and triggering apoptosis. FOXO4-DRI competes with endogenous FOXO4 for p53 binding, freeing p53 to initiate programmed cell death.
Selective Senescent Cell Elimination
Because only senescent cells rely on the FOXO4-p53 survival complex, FOXO4-DRI's mechanism is cell-type selective — healthy, proliferating cells express different survival signals and are unaffected by the peptide.
SASP Reduction
Senescent cells secrete a Senescence-Associated Secretory Phenotype (SASP) of inflammatory cytokines, proteases, and growth factors that damage surrounding tissue. Eliminating these cells reduces the chronic SASP-driven inflammation underlying age-related tissue deterioration.
Tissue Rejuvenation
Following senescent cell clearance, resident stem cell populations are released from SASP-mediated inhibition, enabling tissue repair and regeneration that was suppressed by the senescent cell burden.
